Normal Expression

  • YY1 localization changed during development and that expression of gamma-smooth muscle actin correlated with expression of cytoplasmic rather than nuclear YY1. PMID: 16314465

Abnormal Expression

  • YY1 protein levels are increased in human heart failure and that YY1 represses the activity of the human alphaMyHC promoter. PMID: 15367688

Function

  • YY1, in addition to its regulatory roles in normal biologic processes, may possess the potential to act as an initiator of tumorigenesis and may thus serve as both a diagnostic and prognostic tumor marker; furthermore, it may provide an effective target for antitumor chemotherapy and/or immunotherapy. Applications

 

EMSA

  • YY1 activity, shown by electrophoretic mobility shift assay, decreased upon infection in the nuclear extracts but remained unchanged in whole cell extracts until 48h post-infection (p.i.). Immunohistochemical staining of the fixed cells showed translocation of the factor to the cytoplasm of the infected macrophages. PMID: 15084399

  • Using electrophoretic mobility shift (EMSA) technique, we observed that glutamate treatment induced a dramatic increase in the YY1-binding activity of the smallest of three complexes both in the hippocampal and cerebellar granule cells. PMID: 9495557

  • Highly purified YY1 behaved as authentic YY1 on electrophoretic mobility shift assays and in enhancing transcription from the P6 promoter. PMID: 7606165

  • using electrophoretic mobility shift assay, we have identified YY1-related factors that bind to two other transcription initiators in cellular genes. PMID: 1720509

Western Blotting (WB)

  • Supershift assays with specific YY1 antibody showed that all three complexes contained YY1 protein. Western blot assays did not show any changes in the nuclear levels of YY1 protein. PMID: 9495557
